Özet
Pistachio production relies heavily on the availability of genetically diverse and resilient rootstocks, yet the molecular characterization of interspecific Pistacia hybrids remains largely unexplored. This study provides the first comprehensive simple sequence repeat (SSR)-based assessment of hybrid Pistacia genotypes developed within a national rootstock breeding program in Türkiye. Forty-seven genotypes derived from four species commonly used as rootstocks (P. atlantica, P. integerrima, P. eurycarpa, and P. terebinthus) were genotyped using 38 highly polymorphic SSR markers developed from P. vera. A total of 355 alleles were identified, with an average of 9.34 alleles per locus, reflecting substantial genetic variability across the population. The mean observed and expected heterozygosity values were 0.74 and 0.77, respectively, while the average polymorphism information content (0.74) confirmed the high discriminatory capacity of the SSR markers. Transferability of SSR markers developed from P. vera across to wild Pistacia genotypes were calculated and CUPOhPa590 generated the highest transferability (94%), followed by CUPSiPa124 (92%), CUPOhPa1636 (90%), and CUPOhPa173 (90%) markers. Unweighted Pair Group Method with Arithmetic Mean (UPGMA) clustering separated the Pistacia genotypes into two major groups and genetic dissimilarities among the genotypes were identified in the pairs of accessions. In conclusion, the findings highlight the value of interspecific hybrids as promising genetic resources for genetic diversity assessment, SSR marker utility, hybrid identification and selection of materials for future breeding program.
