Sivas ve Yozgat İllerinde Asma Gövdelerinde Sorun Fungal Hastalılarının Teşhisi ve Yaygınlık Durumlarının Belirlenmesi

Özet

Bu çalışma, Türkiye’nin soğuk ve kurak iklim özelliklerine sahip Sivas ve Yozgat illerinde bağcılık yapılan alanlarda asma gövdelerinde görülen fungal hastalıkların teşhisi, yaygınlığı ve şiddetlerinin belirlenmesi amacıyla yürütülmüştür. Sürvey çalışmalarında, güdümlü örnekleme yöntemiyle belirlenen hastalıklı omçalardan fungal hastalık etmenlerinin izolasyonu ile morfolojik ve moleküler tanımlamaları yapılmıştır. Patojenlerin tanılanması, ITS 4 ve ITS-5 primerleri kullanılarak ITS (Internal Transcribed Spacer) dizilerinin amplifikasyonu ve sekanslanmasıyla, virülenslik düzeyleri ise patojenisite testleriyle belirlenmiştir. İzolasyonlar sonucunda, fungal hastalıkların Yozgat ilinde %75 yaygınlık, %3.1 çıkış ve %8.3 şiddetinde; Sivas ilinde ise %71.4 yaygınlık, %4.1 çıkış ve %15.4 şiddetinde olduğu belirlenmiştir. Yozgat ilinden 6 patojen fungal tür (Botryosphaeria dothidea, Diplodia seriata, Kalmusia variispora, Macrophomina phaseolina, Neofusicoccum parvum, Phaeoacremonium aleophilum); Sivas ilinden ise 3 patojen fungal tür (Diatrype stigma, Kalmusia variispora, Phaeoacremonium minimum) tespit edilmiştir. Patojenisite test sonuçlarına göre en virülent tür Neofusicoccum parvum (21.7 mm) olarak belirlenmiş, Phaeoacremonium minimum (7.2 mm) ve Macrophomina phaseolina (8.7 mm) ise daha düşük virülenslik oranı göstermiştir. Sörvey yapılan bölgelerde asma gövde hastalıklarına neden olan fungal etmenler ilk kez kapsamlı olarak bu çalışma ile ele alınmıştır. Elde edilen sonuçlar, bölgede bağcılığın sürdürülebilirliği açısından gövde hastalıklarının potansiyel tehdit oluşturduğunu ortaya koymakta ve gelecekte bu bölgelerde hastalıklarla mücadele stratejilerine bilimsel temel sağlamaktadır. 

This study aimed to determine the identification, prevalence, and severity of fungal diseases affecting grapevine trunks in vineyards located in the provinces of Sivas and Yozgat, which have cold and arid climates. During the survey, fungal pathogens were isolated from diseased vine shoots identified using a guided sampling method, and their morphological and molecular characteristics were analysed. Pathogen identification was carried out by amplifying and sequencing the ITS (Internal Transcribed Spacer) regions using ITS-4 and ITS-5 primers, while virulence levels were assessed through pathogenicity tests. The results of the isolations showed that fungal diseases had a prevalence of 75%, an incidence of 3.1%, and a severity of 8.3% in Yozgat Province; and a prevalence of 71.4%, an incidence of 4.1%, and a severity of 15.4% in Sivas Province. Six pathogenic fungal species (Botryosphaeria dothidea, Diplodia seriata, Kalmusia variispora, Macrophomina phaseolina, Neofusicoccum parvum, Phaeoacremonium aleophilum) were identified in Yozgat Province, and three pathogenic fungal species (Diatrype stigma, Kalmusia variispora, Phaeoacremonium minimum) were identified in Sivas Province. According to the pathogenicity test results, the most virulent species was Neofusicoccum parvum (21.7 mm), while Phaeoacremonium minimum (7.2 mm) and Macrophomina phaseolina (8.7 mm) exhibited lower virulence. This study is the first comprehensive examination of fungal pathogens causing grapevine trunk diseases in these regions. The results indicate that trunk diseases pose a potential threat to the sustainability of viticulture in the region and provide a scientific basis for future disease control strategies.
