The Effects of Different Cytokinins on Micropropagation of 'Garnem' Rootstock

Abstract

In this study, the propagation ability of 'Garnem' rootstock which is a clonal almond x peach hybrid (G x N15) was investigated by shoot tip in vitro culture. Apical and axillary buds of shoots were used as explant material. MS medium containing different concentrations of BAP (0.0, 1.0, 2.0, 4.0 mg/L), Kin (0.0, 1.0, 2.0, 4.0 mg/L), 2iP (0.0, 1.0, 2.0, 4.0 mg/L), and TDZ (0.0, 0.1, 0,2, 0,4 mg/L) was tested for propagation. The cultures were incubated in culture room at 24 +/- 2 degrees C with a 16-h period under 3000-4000 lux light irradiance. The effects of PGR on multiplication rate of 'Garnem' rootstock were determined. Considerable differences were found in response to hormone concentrations. The best number of shoot was found from the media containing 1 mg/L BAP. Higher BAP (2.0, 4.0 mg/L) concentrations, decreased the coefficient of shoot.
