THE EFFECTS ON HEMATOLOGICAL PARAMETERS OF Yersinia ruckeri INFECTION in TROUTS (Oncorhynchus mykiss) FED WITH ROSEHIP (Rosa canina)

Özet

Objective: This work was conducted as experimental in order to investigate some hematological changes as a result of Yersinia ruckeri infection on trouts (Oncorhynchus mykiss) which their feeds were added different rates (10%, 20%, 30%) of rosehips (Rosa canina).

Methods: Experimental fish were got from a trout fish farm. Then have been put in 10 round fiber tank with 0,25 m3 in Fish Diseases Unit of Fisheries Faculty. The work has been organized as 3-repeated. Experimental groups which were fed 10% (1st group), 20% (2nd group), 30% (3rd group) rates of rosehip (Rosa canina) containing feed, positive (4th group) and negative (5th group) control groups. It consisted of total of 150 fish. To create experimental infection has been designated 3×108 cfu/ml Yersinia ruckeri inoculum as the appropriate dose. In 8 tank located in group of 15, for a total of 120 fish have been injected 0,1 ml as intraperitonally. On fifth day after  the infection were got 10 sample from control and experimental groups. They were examined as hematological. The measurement of leukocytes (WBC), lymphocytes, monocytes, granulocytes, erythrocyte (RBC), erythrocyte indexes (MCV, MCH, MCHC), hematocrit (Hct) and hemoglobin (Hb) in blood parameters have been measured with "MS4-e Veterinary Hematology Analyser (Hemocell counter)" in Trout Fishing Scales.

 Statistical analysis of results, control and experimental groups have been tested with One-Way Anova and Duncan in SPSS Statistical Program. The differences experimental groups and levels of importance of these differences have been determined (P <0.05 P> 0.05).

Results and Discussion: Hct and RBC measurement results 1st group was a decrease compared with 2nd, 3rd and 4th group. But 5th group was an increase. There was no change in the number of WBC between control and experimental groups (P <0.05). The result of lymphocyte percentage 5th group was decrease compared to other groups Monocytes in 1st group was determined a reduction compared to other groups (2/3/4 and 5th groups) (P <0.05). The results of granulocytes and Hb,  5th group was detected an increase compared to other groups (1/2/3 and 4th group). In terms of MCV 2nd group was determined a decrease compared to other groups (1/3/4 and 5th group). The result of MCHC and  MCH 2nd group were decrease but 3rd and 1st groups were increase compared to 4th and 5th groups (P <0.05)

According to the results of our research, there were a significant reductions in erythrocyte and hemoglobin amounts depending on bacterial agents in septicemic disease. In addition, feeding with rosehip was stimulate to  lymphocyte cells against bacterial agent.
