Investigation of Viroid Diseases in Lemon Trees with Rumple and the Distribution of Rumple Disease in Lemon Growing Areas of Erdemli District in Turkiye

Abstract

The purpose of this study was to determine the frequency of Rumple disease in lemon (Citrus lemon L.)  growing regions of the Erdemli district of Mersin Province in Turkiye as well as to identify citrus viroid agents in the diseased trees. Surveys on the Kütdiken, Meyer, Aydın, and Interdonato lemon cultivars were carried out in the local lemon orchards. Rumple disease had a cumulative prevalence rate of 29% in the Erdemli district's lemon-growing areas. The major symptoms seen in biological indexing tests were epinasty and leaf necrosis on leaves as a result of mechanical inoculation on etrog citron seedlings grown from seed. To find out if citrus exocortis viroid (CEVd) and citrus cachexia viroid (CCaVd) were present in trees exhibiting collapse symptoms, RT-PCR tests were conducted using samples from 10 different lemon trees. Following the studies, it was found that 6 of the samples were infected with CEVd and had a band at a level of 371 bp. Furthermore, 6 of the tested samples showed CCaVd infection and revealed a band at the 300 bp level.
